چکیده

We observed the nonenzymatic formation of guanosine 3’:5’-monophosphate (cyclic GMP) from GTP. The reaction was dependent upon heat and was accelerated by divalent cations and creatine phosphate. The formation of cyclic GMP was greater with increased incubation time, temperature, GTP concentration, and pH. All of the divalent cations tested (Mg2+, Mnz+, Ca2+, Ba2+, and Znzf) increased cyclic GMP formation. The stimulatory effect of all of the cations tested, except Zn2+, was increased with creatine phosphate. Cyclic GMP formed was determined with two different assay methods and characterized with cyclic nucleotide phosphodiesterase treatment and two different chromatographic systems. It is concluded from our studies that significant amounts of cyclic GMP can be formed nonenzymatically from GTP when guanylate cyclase reaction mixtures are heated at a pH near neutrality.

The Activity of Oligonucleotides Containing Guanosine S-Triphosphate in Protein Synthesis I. THE INTERACTION OF PROTEIN SYNTHESIS ELONGATION FACTOR I WITH CYTIDYLYL (5’-3’)- GUANOSINE 5’-TRIPHOSPHATE*

The interaction of protein synthesis elongation factor 1 (EF-1) from wheat embryos and elongation factor Tu from Escherichia coli with cytidylyl(5’-3’)guanosine 5’-triphosphate(pppGpC) has been studied. The dinucleotide 5’-triphosphate interacts strongly with EF-1 as evidenced by its capacity to inhibit the binding of [3H]GTP to the factor. The analogs pGpC and GpC do not interfere with GTP bin...

Antinociceptive Effect of Vardenafil on Carrageenan-Induced Hyperalgesia in Rat: Involvement of Nitric Oxide/Cyclic Guanosine monophosphate/ Calcium Channels Pathway

In this study, we aimed to investigate the peripheral antinociception effects of specificphosphodiesterase 5 (PDE-5) inhibitor vardenafil on carrageenan-induced nociception in rats, and the role of calcium besides the L-arginine- nitric oxide (NO)- cyclic guanosine monophophate(cGMP) pathway in these effects. Hyperalgesia was induced by the intraplantar injection of 0.1mL fresh carrageenan solut...

Interferon- Gamma- Inducible Guanosine Triphosphate Cyclohydrolase 1 (GTP-CH1) Pathway Is Associated with Frailty in Egyptian Elderly

Background: Chronic low-grade inflammation may be a cardinal pathophysiologic feature in the pathogenesis of frailty. Interferon-gamma (INF-γ) is an understudied proinflammatory cytokine in frailty that induces many inflammatory pathways including the guanosine triphosphate cyclohydrolase 1 (GTP-CH1) pathway. Our aim was to evaluate the GTP-CH1 pathway in Egyptian frail elderly subjects. ...

Phosphorylation of nucleosides and nucleotides with inorganic monoimido-cyclo-triphosphate.

The phosphorylation of nucleosides (adenosine, guanosine, cytidine, and uridine) and nucleotides (adenosine 5'-monophosphate, guanosine 5'-monophosphate, cytidine 5'-monophosphate and uridine 5'-monophosphate) has been achieved using inorganic monoimido-cyclo-triphosphate (MCTP, Na(3)P(3)O(8)NH) in aqueous solution. In this reaction, the 2'-OH or 3'-OH group of the beta-D-ribofuranose unit was ...
